چکیده

The high neuroactive potential of metabolites produced by gut microbes has gained traction over the last few years, with metagenomic-based studies suggesting an important role microbiota-derived γ-aminobutyric acid (GABA) in modulating mental health. Emerging evidence revealed presence glutamate decarboxylase (GAD)-encoding gene, a key enzyme to produce GABA, prominent human intestinal genus Bacteroides . Here, we investigated GABA production culture and metabolic assays combined comparative genomics phylogenetics. A total 961 genomes were analyzed silico 17 metabolically genetically diverse isolates representing 11 species screened vitro Using model organism thetaiotaomicron DSM 2079, determined kinetics, its impact on milieu pH, assessed mitigating acid-induced cellular damage. We showed that GAD-system consists at least four highly conserved genes encoding GAD, glutaminase, glutamate/GABA antiporter, potassium channel. demonstrated prevalence among 90% all (96% only) harboring 16 strains producing (ranging from 0.09 60.84 mM). identified glutamine as precursors production, is regulated acts protective mechanism against stress , cell death preserving activity. Our data also indicate might represent only amino acid-dependent tolerance system Altogether, our results suggest contribution regulation GABAergic gut.

P146: Gamma Aminobutyric Acid (GABA) and its Alterations in Stress

Gamma aminobutyrate (GABA) is a non-protein amino acid that is thought to play an important role in the modulation of the central response to stress. Mechanisms by which GABA may facilitate these responses to stress are metabolic and/or mechanical disruptions. Environmental stresses increase GABA accumulation through cytosolic acidification, induce an acidic pH-dependent activation of glutamate...

Production of phenylacetic acid by strains of Bacteroides asaccharolyticus and Bacteroides gingivalis (sp. nov.).

Strains of Bacteroides asaccharolyticus and Bacteroides melaninogenicus subspecies isolated from human and animal sources were examined for the production of phenylacetic acid. B. asaccharolyticus strains isolated from sites in humans and monkeys always produced phenylacetic acid. B. asaccharolyticus strains isolated from human nonoral sites consistently failed to produce this product. This met...

Black-pigmented Bacteroides spp. in human apical periodontitis.

The incidence of black-pigmented (BP) Bacteroides spp. in 62 human dental root canal infections (35 acute and 27 clinically asymptomatic cases of apical periodontitis) in 57 adults was studied. Altogether 37 strains of BP Bacteroides were found in 31 infections, always in mixed anaerobic infections. Two different BP Bacteroides species were present in six infections. B. intermedius was most fre...
